Pozycja Hepatoprotective effect of Costus afer (Lin) on toxic metal mixture treated rats mediated by regulation of oxidative stress markers, inflammatory cytokines and bio-metal chelation(Rzeszów University Press, 2025-03) Anyanwu, Brilliance O.; Ezejiofor, Anthonet N.; Ajibo, Doris N.; Orisakwe, Orish E.Introduction and aim. Cadmium, lead, and mercury have been considered to exhibit their hepatotoxic effects by oxida tive induction damage and the generation of reactive oxygen species (ROS). The current work evaluated the protective activ ity of aqueous leaf extracts of Costus afer (ALECA) on liver damage arising from exposure to toxic metal mixture (TMM): 1.61 mg/kg cadmium chloride (CdCl2), 20 mg/kg lead chloride (PbCl2), and 0.40 mg/kg mercury chloride (HgCl2). Material and methods. Five groups of weight-matched Sprague Dawley rats were treated for 90 days. Metal mixtures and deionized water were used to treat the 2 groups of rats whereas the other 3 groups were treated with various doses of the ALECA through oral gavage with TMM. Hepatic function parameters, oxidative biomarkers, inflammatory cytokines, mor phological changes, and metal levels in the liver were monitored. Results. Treatment with TMM resulted in significant increases in alanine transaminase, aspartate transaminase, alkaline phos phatase, bilirubin, interleukin 6, malondialdehyde, but decreased albumin, total protein, interleukin 10, superoxide dismutase, catalase, and glutathione levels. TMM also caused some morphological changes and increased the concentrations of heavy metals (Pb, Cd, and Hg) in the liver. Conclusion. ALECA showed beneficial effects against TMM-induced hepatotoxicity via metal chelation, anti-inflammatory, and antioxidant mechanism. With the increased awareness from circular bioeconomy that focuses on ‘no waste’ generation mantra, various technologies have been developed to valorize these wastes into useful products, including melon seed husk. The aim of the study was to evaluate the effects of silica nanoparticles from melon seed husk (SiNPs MSH) against Ni, Al, and Ni/Al mixture-induced hematotoxicity and lipotoxicity in Sprague Dawley rats. Material and methods. Fifty-six male Sprague Dawley, 6 to 8 weeks and weighing 220 to 250 g, were randomly allocated to eight groups (n=7). Group 1 received deionized water only (control), groups 2, 3 and 4 (exposed groups) received the Ni/Al mixture, 0.2 mg/kg Ni and 1.0 mg/kg Al, while groups 5 to 8 received the Ni/Al mixture, Ni, and Al plus 100, 200 and 400 mg/kg of SiNPs respectively for 90 days. Blood samples were collected for biochemical investigation. Results. Ni, Al and Ni/Al groups showed significant (p<0.05) alteration (p <0.05) in the classic lipid profile, hematological and oxidative stress markers compared to the control. Co-administration with SiNPs did not show significant (p>0.05) difference in these parameters compared to the control. Conclusion. MSH SiNPs reversed Ni, Al, and Ni/Al mixture mediated hemotoxicity and elevated superoxide dismutase, catalase, and glutathione probably via metal chelation.Pozycja Zn and Se supplementation abrogated metals-(metaloids) mixture mediated ocular-thymus toxicity via modulation of oxido-inflammatory and antiapoptotic mechanisms in female Sprague Dawley rats(Publishing Office of the University of Rzeszow, 2024-12) Antia, Mfoniso; Ezejiofor, Anthonet N.; Orish, Chinna N.; Ugwu, Theresa; Cirovic, Ana; Cirovic, Aleksandar; Ajibo, Doris N.; Orisakwe, Orish E.Introduction and aim. This is an evaluation of the protective effects of Zn and Se in the eye and thymus of rats exposed to cock tail noxious metal mixtures (CNMM) (Al, Pb, Hg and Mn) in ameliorating ocular pathologies due to autoimmunity. Material and methods. Female Sprague rats were grouped into eight (n=5) and orally exposed to various treatments for a period of 60 days: (1): the control group receive deionized water only; (2): the CNMM only group received lead acetate Pb(C2H3O2)2 (20 mg/kg), AlCl3 (35 mg/kg), HgCl2 (0.40 mg/kg) and MnCl2 (0.56 mg/kg); (3) received CNMM+ZnCl2, 0.80 mg/kg; (4) received CNMM+Na2SeO3, 1.50 mg/kg; (5) received CNMM+ZnCl2, 0.80 mg/kg and Na2SeO3, 1.50 mg/kg combination. Oxidative stress markers, nuclear factor erythroid 2-related factor 2, nuclear factor kappa B, interleukin 6, tumor necrosis factor alpha and caspase-3 and histopathological changes were determined. Results. CNMM decreased antioxidants levels but increased malondialdehyde and nitric oxide concentrations. CNMM in creased levels of nuclear factor erythroid 2- related factor 2, and nuclear factor kappa B, interleukin 6 and tumor necrosis factor alpha and caspase-3. There was moderate retinal degeneration and total cell loss at the ganglionic cell layer in the eye; severe degenerative thymus, lymphocyte depletion and multifocal necrosis in CNMM only. Conclusion. Supplementation with Zn and Se reduced the biochemical and histopathological changes in the eye and thymus in response to CNMM exposure.
