Mapowanie genów odporności na mączniaka prawdziwego u odmian pszenżyta ozimego ‘Lamberto’ i ‘Grenado’

Abstrakt
Pszenżyto (X Triticosecale Wittmack) jest stabilnym amfiploidem, powstałym w wyniku międzyrodzajowej hybrydyzacji, po której następuje podwojenie chromosomów, formy matecznej gatunku Triticum (AABB) oraz formy ojcowskiej gatunku Secale (RR). Przez wiele lat pszenżyto było uważane za zboże mało podatne na choroby, jednak w ostatnim 10-leciu nasilenie chorób na pszenżycie zwiększało się. W 2004 roku nastąpiło załamanie odporności pszenżyta na mączniaka, szczególnie u odmiany ‘Lamberto’. Celowym i ekonomicznym sposobem ochrony pszenżyta przed chorobami powodowanymi przez grzyby patogeniczne jest uprawa odmian odpornych. Wykorzystanie narzędzi biologii molekularnej, w tym markerów DNA, umożliwia poznanie genetycznego podłoża odporności pszenżyta na infekcję Blumeria graminis, przez co wspomaga selekcję odmian odpornych. Konstrukcja map genetycznych oraz identyfikacja loci cech ilościowych pozwala na lokalizację potencjalnych miejsc w genomie powiązanych z daną cechą. W niniejszej pracy mapę genetyczną pszenżyta skonstruowano dla dwóch populacji powstałych poprzez skrzyżowanie odmian ‘Lamberto’ (wrażliwa) oraz ‘Grenado’ (odporna) w dwóch kierunkach. Wykorzystano systemy markerowe DArT oraz SSR. Ocena fenotypowa, przeprowadzona w stadium siewki oraz rośliny dojrzałej, oparta była na ocenie stopnia porażenia populacji mapującej patogennym grzybem – Blumeria graminis. W celu zidentyfikowania rejonów/markerów genomu związanych z odpornością na infekcję wykorzystano analizy: Kruskal-Wallisa, SMA (dla oceny inokulacyjnej) oraz dodatkowo IM oraz CIM (dla oceny polowej). Wynikiem pracy jest mapa genetyczna dwukierunkowej populacji pszenżyta ozimego, o długości 1 813.6 cM, złożona z 547 markerów DArT oraz 7 markerów SSR. Zidentyfikowane zostały translokacje niehomeologiczne 5BS.3BL oraz 7A.1A. W stadium rośliny dojrzałej zidentyfikowano dwa główne efekty związane z odpornością na Blumeria graminis – Q.Pm.lgl.1R oraz Q.Pm.lgl.6R, wyjaśniające odpowiednio 15% oraz 33.4%. Do istotnych efektów należą także Q.Pm.lgl.2A2, Q.Pm.lgl.3A oraz Q.Pm.lgl.1B. U roślin w stadium siewki zidentyfikowano markery sprzężone z cechą w grupach sprzężeń 1R oraz 6R. Wytypowano markery znajdujące się w sąsiedztwie opisanych markerów sprzężonych z genami Pm50 i Pm4 na chromosomie 2A, oraz Pm39 na chromosomie 1B. Wskazuje to na możliwe występowanie w dwukierunkowej populacji ‘Lamberto’ i ‘Grenado’ w/w genów. Uzyskane wyniki dostarczyły nowych danych na temat wpływu kierunku krzyżowania odmian pszenżyta na dziedziczenie i/lub ekspresję genów. Informacje dotyczące podłoża genetycznego odporności, w tym rejonów genomu populacji ‘Lamberto’ i ‘Grenado’ związanych z odpornością na Blumeria graminis, mogą stanowić podstawę do opracowania wydajnego systemu selekcyjnego hodowli roślin odpornych.
Triticale (X Triticosecale Wittmack) is an amphiploid stably carrying the genomes of wheat (AABB or AABBDD) and rye (RR). Triticales are the fertile progenies of an intergeneric hybridization between a female parent from the genus Triticum and male parent from the genus Secale, followed by chromosome doubling. The economic importance of triticale systematically grows thanks to its better performance in less favorable environments. In recent years, resistance to pathogenic fungus – Blumeria graminis, that causes powdery mildew, significantly decreased in triticale. Deliberate and economical way to protect triticale against diseases caused by pathogenic fungi is the cultivation of resistant varieties. The identification of natural sources of resistance and breeding resistant varieties is the most effective way to control powdery mildew disease. Phenotypic selection based on molecular markers linked to genes for resistance can also facilitate resistance breeding. All triticale breeding programs require saturated genetic maps to locate quantitative traits of resistance. Genetic map construction and analysis of quantitative trait loci allows for the location of potential sites in the genome associated with the interesting trait. In the present study, bidirectional crosses between ‘Lamberto’ and ‘Grenado’ as susceptible and resistant parent, respectively, were developed in order to obtain F2 and F3 mapping populations. Genetic linkage maps based on genotyping of ‘Lamberto’ × ‘Grenado’ (LG) and ‘Grenado’ × ‘Lamberto’ (GL) populations with DArT and SSR markers were constructed. Both populations were inoculated and evaluated in controlled conditions and field tests. QTLs were identified by single marker analysis, interval mapping, composite interval mapping analyses and validated by Kruskal-Wallis test. The result of this study is a genetic map of winter triticale of total length 1 813.6 cM with 554 loci (547 DArT markers and 7 SSR markers). Translocations of chromosomes fragments from different homeologic groups (5BS.3BL and 7A.1A) were identified. Two main, stable QTLs on chromosomes 1R (Q.Pm.lgl.1R) and 6R (Q.Pm.lgl.6R) were identified, explaining 15% and 33.4% of variation, respectively. Three DArT markers mapped on 6R chromosome (rPt-398825, tPt-513060, rPt-411235) were associated with resistance to powdery mildew. Minor effects in adult plant stage were also identified in 2A2, 3A and 1B linkage groups. The possible occurrence of powdery mildew resistance genes: Pm4, Pm 50 (on chromosome 2A) and Pm39 (on chromosome 1B) was discussed. Genomic regions/markers of the bidirectional population ‘Lamberto’ and ‘Grenado’ associated with resistance to Blumeria graminis constitute a basis for development of efficient selection system.
